Palonosetron is an anti emetic agent and a drug used in nausea. It belongs to the group of serotonin or 5HT3 receptor agonist which acts on the various locations having this receptor such as the GIT and CTZ to prevent the emetic stimuli from working. The drug is available in injectable form and is water soluble and can be given in various conditions which can induce vomiting in people.


Palonosetron HCl is used as an anti-emetic injection in case of patients who are being given anticancer drugs. The drug for preventing vomiting is possible to be given in the initial as well as during the repeat doses of anticancer therapy. Since, this kind of vomiting is not subsided easily by any other antiemetic drug, Palonosetron is being used nowadays effectively. Although, this drug can be used for any other kind of emesis prevention and treatment, it is mostly being used for the above purpose.

Side effects

During the clinical trials, a few side effects of the drug have been seen. In the CVS, the drug may cause tachycardia or bradycardia, hypotension, arrhythmia, and some other effects. Allergic reactions, rashes, tinnitus, ambylopia, dyspepsia, dry mouth, hiccups, flatulence, flu like syndrome, increase in serum liver enzymes, and some metabolic syndromes are to be taken care of as these can sometimes lead to the stoppage of the drug treatment.


Palonosetron HCl is not to be administered in patients who are previously allergic to the drug or any of its components.


The drug vial is to be stored in temperatures of around 25 deg C and within a range of about 15-30 degree C. They should be protected from freezing and from light and from the reach of children.


The aloxi injection contains a nonpyrogenic, isotonic buffered solution for intravenous administration. Each 5 ml vial injection contains 0.25 mg or 250 mcg of Palonosetron base as hydrochloride along with other preservatives and agents such as disodium adetate, mannitol, etc in water for giving intravenously.

Important Precautions

Hypersensitivity to the drug in any previous occasion should be made known to the doctor. Cardiac disturbances of the type in which there is precipitation of arrhythmia are to be noted, as the drug can interact with other drugs which can cause QT prolongation. Since the effects of the drug on the mutagenesis and carcinogenesis are not yet established, their use in pregnant females is not to be done. Therefore, females are advised not to get pregnant if they are on this drug or should stop the drug immediately if they are planning for pregnancy. During the treatment process, if there are reports of hepatic and renal impairment, they should be immediately stopped.

Dosage and administration

The drug is given in the dose of 250 mcg in a 5 ml vial which is injected intravenously over a period of about 30 seconds. It should be given about 30 minutes before the chemotherapy is started. The drug is not to be mixed with any other medications and therefore a new IV line set is used or the old one is flushed with normal saline before using this drug.
