Introduction

Rasagiline mesylate is a drug belonging to the group of selective MAO-B inhibitors which is nowadays being used for the therapy of Alzheimer's disease. By inhibiting the monoamine oxidase enzyme type-B, present mostly in the brain, the drug is able to allow the action of serotonin and other neurotransmitters to act for a longer period of time. This helps in improving the symptoms of alzheimers disease and provides symptomatic relief. This is an orally administered drug that is well absorbed from the intestine and reaches the brain to act in the CNS.


Use

Due to the MAO-B inhibitory action of Rasagiline, it can be used in the alzheimers disease conditions, either as single drug or as an adjunctive therapy to levodopa which is the primary drug for the condition. Studies have shown that this drug is more efficacious and has a better effect on the symptoms when used as an adjunct.


Side effects

The side effects of the drug are not very severe. But when administered as an adjunct with other drugs, the number of problems increases. Common side effects that are seen with Rasagiline mesylate are headache, arthralgia, dyspepsia, flu syndrome, conjunctivitis, malaise, etc, which are not serious enough to withdraw the drug. Allergic reactions can occur rarely while other rare effects can be alopecia, angina pectoris, hallucinations, etc.


Contraindication

The drug is contraindicated to be used along with Meperidine and other MAO-B inhibitors and this condition is termed as serotonin syndrome. It should also be not given along with drugs belonging to analgesic category. It is also dangerous to give the drug to patients who have allergy to Rasagilione or any other MAO-B inhibitor.

The drug should be usually stored at temperature of about 25 °C and should be within the range of 15-30°C.


Composition

The drug Azilect tablet contains 1 mg equivalent of Rasagiline base in form of Rasagiline mesylate. The drug also contains binders and preservatives such as mannitol, starch, colloidal silicon dioxide, stearic acid, etc.


Important Precautions

The drug should be avoided being used with other MAO-B inhibitors and this information should be given to the prescribing doctor. According, the dose can be changed or the other drugs can be stopped. Hepatic impairment and renal impairment conditions should have reduction of the dose of Rasagiline mesylate as it gets metabolised in the liver and removed through the kidney. Due to the risk of hypertensive crisis when used with other inhibitors at higher doses, the dose should never be changed without the consultation of the physicians.


Dosage and administration

Rasagiline mesylate is administered as tablets of 1 mg once daily when used as a monotherapy. When the use is as an adjunct, it can be given as 0.5 mg tablet and the dose can be slowly increased to 1 mg once daily. It is available as round tablets with bottles containing 30 tablets in total.
