This thiocarbamide compound has been proved effective in managing cases of hyperthyroidism usually seen in Grave's disease. This anti thyroid agent decreases the production of triiodo and tetraiodothyroxine level which are profusely secreted during the disease. It inhibits the peroxidise enzyme and also prevents iodine from binding to the thryoglobulin and thus terminates the constant synthesis of the thyroid hormone. The drug also prevents the conversion of T4 to its active form T3 and lowers the activity of thyroid hormone.


The drug propylthiouracil is indicated in cases of overactive thyroid gland where it lowers the level of the active form of the hormone. It is also used in decreasing the symptoms of hyperthyroidism in preparations for surgically removing the thyroid gland. The drug can also be used to treat cases of hyperthyroidism when it is not advisable to remove the gland.

Side effects

The most common side effects encountered in the use of the drug are nausea, vomiting, headache, GI disturbances, arthralgia, fever, urticaria, skin rash, thrombocytopenia, cutaneous vasculitis and lupus like syndrome. Fatal side effects like agranulocytosis, aplastic anaemia, liver injury and liver failure has also been encountered.


The drug is contraindicated in children less than 6 years of age. The drug should not be given to patients suffering from previous history of allergic reactions such as blood or skin disorders. Pregnant and lactating females should not be prescribed with the drug.


The drug propylthiouracil should be stored in a temperature ranging from 15 to 35 degree Celsius. It should be kept away from direct heat and sunlight. The product should be kept in a place away from reach of children.


Each tablet contains Propylthiouracil 50 mg and other active ingredients like anhydrous lactose, magnesium stearate, microcrystalline cellulose, pregelatinized starch, and sodium starch glycolate.

Important Precautions

Propylthiouracil should be cautiously used in patients suffering from liver and renal impairment and those with past history of allergic reactions. The treatment should be stopped immediately if any type of liver dysfunction is observed. Regular monitoring of thyroid function and CBC is advisable. The prothrombin time should be monitored regularly during the treatment and before any surgical procedures. Any symptoms like agranulocytosis, fever and aplastic anaemia should follow immediate stoppage of the treatment.

Dosage and administration

The drug should be initiated with a dose of 300 mg/day. In severe cases the dosage may exceed 300 mg/day. It should be taken every 8 hours. The long term adult dosage after the initial treatment should remain between 100 to 150 mg/day. The drug should always be taken with food.
